Nitrous oxide emissions from soil are shaped by both soil chemical properties and microbial community composition, but the relative impact of each factor is not fully understood. In this study, we extracted microbiomes from two distinct soil types, black soil and fluvo-aquic soil and inoculated into soil water extracts to investigate how water-extractable chemicals and microbiomes independently affect Nitrous oxide emissions. A total of 108 DNA samples (triplicates per treatment) were collected from 12 treatment groups across three time points (D0, D2, and D7). The V3-V4 region of the 16S rRNA gene was amplified using universal primers (341F and 785R) for Illumina MiSeq sequencing, following standard protocols (Illumina Inc., USA).
