To identify the gene expression changes that depend on the activity of CIC (Capicua), we treated Kras(+/G12V);Trp53(-/-) or Kras(+G12V);Trp53(-/-);Cic(D2-6/D-2-6) lung tumor cells with trametinib to block MEK which will activate the CIC repressor. We predicted that CIC activation in Kras(+/G12V);Trp53(-/-) cell would cause repression of genes controlled by CIC. In contrast, Kras(+G12V);Trp53(-/-);Cic(D2-6/D-2-6) lung tumor cell lines express inactive CIC proteins and treatment with trametinib would not cause repression of CIC target genes.
