We determined the cell types present in Arabidopsis callus by single-cell RNA-seq with Quartz-Seq2. Overall design: We dissociated Arabidopsis callus generated from hypocotyl explants to single-cell by enzymatic treatment. Single cell was sorted into lysis buffer in each well of a 384-well PCR plate using BD FACS Aria II SORP cell sorter or CellenOne. Sequence library DNA was prepared according to Quartz-Seq2 method. We analyzed the sequence library DNA using NextSeq500. Digital expression matrix was calculated with fastq files.
