The heterogeneous characteristics of activated monocytes are much unknown. We performed the single cell transcriptomic analysis of LPS-treated human monocytes from healthy blood donor, and CD127 expression associated expression pattern of inflammatory genes was identified in LPS-treated monocytes. Overall design: PBMC were isolated from buffy coat of healthy blood donor by Ficoll gradients, and CD14+ monocytes were isolated from PBMC with anti-CD14 magnetic beads (Miltenyi Biotec). CD14+ monocytes were treated with LPS for 6 h and then frozen for preservation to sent out for scRNA-seq library construction by using 10x genomics platform.
