This study on blastoderm stage Drosophila embryos seeks to measure accessible chromatin and the enrichment on chromatin of transcription factors associated with BMP signaling (phospho-Mad, Medea) in either wild-type conditions, or in embryos mutant for the homeodomain transcription factor zerknullt (zen). The associated manuscript demonstrates that zen is a critical transcription co-activator that enables rapid responses to BMP signaling. Overall design: Two measurement types were performed. 1) Chromatin IP (ChIP) was performed for either phospho-Mad or GFP-tagged Medea (or negative controls) in wild type embryos. GFP-tagged Medea was ChIPped in zen mutant embryos. 2) Chromatin accessibility was measured by ATAC-seq on single wild-type or zen mutant blastoderm stage embryos.
