BioNANO sequencing of winged bean using Saphyr chip

Ultra-high molecular weight (UHMW) genomic DNA was extracted from plant tissue using the Bionano Prep™ Plant Tissue DNA Kit (Bionano Genomics, Cat. No. 30068), following the manufacturer’s protocol. For optical mapping, the Direct Label and Stain (DLS) protocol was employed, wherein 750 ng of UHMW DNA was labeled using the Bionano Prep DLS DNA Labeling Kit (Bionano Genomics, Cat. No. 80005) according to the manufacturer’s instructions. Following labeling, the DNA was purified and loaded onto a Saphyr chip, and imaging was performed using the Saphyr optical mapping instrument (Nucleome Informatics Pvt. Ltd., Hyderabad, India).
