Chenopodium ambrosioides is a perennial herb native to tropical America and is reported to be a Mn/lead (Pb) hyperaccumulator. In China, C. ambrosioides is well known as a common invasive alien species and can flourish in HM-contaminated habitats. In our previous study, we assessed the genetic and epigenetic variation of 21 C. ambrosioides populations in China, which covered a wide geographical distribution and different HM-contaminated habitats. Two different genetic clusters were detected in the eastern and southwestern populations. We also found that epigenetic variation might contribute to local metalliferous habitat adaptation. In this study, we assessed the molecular basis of three different Mn-tolerant C. ambrosioides populations. Among them, XC and CS belong to one genetic cluster and are distributed in Mn-contaminated and uncontaminated habitats, respectively, while WH belongs to the other cluster and is distributed in uncontaminated habitats. By integrating physiological, transcriptomic and epigenomic data, we investigated i) the differences in transcriptional and physiological responses to Mn stress among three C. ambrosioides populations and ii) the effect of epigenetic variation on Mn tolerance in C. ambrosioides.
