To evaluate the biological effects at the transcriptional level of manufactured nanomaterials, whole genome microarrays of rat alveolar macrophages (NR8383) exposed to three types of submicron-diameter carbon fibers (SCFs) with different physicochemical properties were performed. In comparison with the fibers of SCF1, the fibers of SCF2 were shorter, and the fibers of SCF3 were thinner. Rat alveolar macrophages (NR8383) were exposed to 10 µg/mL of three kinds of SCFs (SCF1, SCF2, and SCF3) for 24 hours, respectively. The experiments were performed using vehicle control (n=4). Cyanine-3-labeled cRNA prepared from NR8383 exposure to SCFs was hybridized to a Whole Rat Genome Microarray. The scanned images of hybridized microarray slides were analyzed numerically to evaluate the biological effects at the transcriptional level of SCFs on NR8383.
