T cell transcriptome after immunofilament or dendritic cell stimulation

In this study we developed immunofilaments - nanosized synthetic polymers - for the antigen-specific activation of T cells. Immunofilaments effectively stimulate CD8+ T cells, but it is unclear how well this stimulation compares to natural dendritic cell activation. In this study, we want to investigate the pathways triggered by the immunofilament and the dendritic cell. RNA-seq profiling of sorted viable CD8+ OT-I T cells after different timepoints (0 hrs (only unstimulated), 8 hrs and 22 hrs) of stimulation. Cells were stimulated with either dendritic cells pulsed with the SIITFEKL-antigen or immunofilaments decorated with SIITFEKL loaded major histocompatibility complex (MHC) and Interleukin-2 (IL-2). Similarities and differences after the stimulation with either immunofilaments or dendritic cells were obtained by comparing to the 0 hr unstimulated timepoint.
