PELP1/SRC-3-dependent regulation of metabolic kinases drives therapy resistant ER+ breast cancer [3D]

Trascriptome analysis of mcf7 cell lines were performed MCF-7 PELP1 cells expressing LXSN, WT PELP1, or cytoplasmic (cyto) PELP1 were used to create tumorsphere 3D culture: Single cells were plated serum-free mammary epithelial basal medium containing 1% methylcellulose, 1% B27 supplement, 1% penicillin–streptomycin, 5 mg/mL insulin, 20 ng/mL EGF, 1 ng/mL hydrocortisone, and 100 mmol/L beta-mercaptoethanol in ultra-low attachment plates and cultured for 6 days prior to RNA isolation.
