Cancer-cell-derived cGAMP limits the activity of tumor-associated CD8+ T cells

To evaluate how CD8+ T-cells are influenced by their STING expression within tumors, we performed single-cell RNA sequencing of reisolated CD8+ T-cells sorted from LLC-cGAStetON-OVA tumors. Overall design: 3 days after tumor (LLCcGAStetON) injection, STING-expressing and STING-deficient OT-I CD8+ T cells adoptively cotransferred to the mice. 6 days after transfer CD8+ T cells were reisolated by flow cytometric cell sorting (live, CD8+, CD45.1/1+ (STING-deficient) or CD45.1/2+ (STING-expressing)), and subjected to single-cell RNA sequencing.
