Single-cell transcriptomic profiling of Botrylloides diegensis of a whole colony

In this study, we performed single-cell RNA sequencing (scRNA-seq) of an entire B. diegensis colony, including zooids, buds, and vascular tunics, to resolve cellular heterogeneity and identify cell and tissue markers. Colonies were fixed in a mix of methanol and acetic acid (ACME), filtered, FACS sorted and loaded on a 10X Chromium Next GEM Single Cell 3ʹ v3 Kit.
