This dataset contains 3D traction force microscopy images of single cells and small clusters of induced pluripotent stem cell-derived endothelial progenitor cells (iPSC-EPs) cultured for 4 or 7 days. iPSC-EPs were embedded in three types of norbornene-functionalized hyaluronic acid-based hydrogels, which were prepared with different concentrations of enzymatically degradable peptide crosslinker resulting in cross-linking levels of 25%, 50% and 75% which correspond to approximate stiffnesses of 190 Pa, 336 Pa, and 551 Pa, respectively. Matched images were acquired in both basal conditions and after treatment with the actin polymerization inhibitor, cytochalasin-D. The iPSC-EPs stably expressed tdTomato (red channel), the nuclei were stained with Hoechst (blue channel), and the hydrogels contained 0.5 µm Dragon Green fluorescent beads (green channel). Each channel provides image data corresponding to cell morphology, cell count, and hydrogel displacement, respectively. Data was collected on a Nikon AXR confocal microscope equipped with a heated chamber set to 37 C, a Queensgate piezo stage, and NIS Elements 6.02.03. Files can be opened with that software, NIS Elements Viewer 5.21.00, Fiji/ImageJ containing the Bio-formats 4.0.0 library, or higher versions.
