combination of culture dependent and culture independent methods of microbial characterization had been done on extremely polluted Adyar river . culture dependent method is by means of separating by growing in artificial media following by the colony morphology, gram staining and biochemical assay with the input of ABIS software.The culture independent method was done by amplification with V3 region of 16 s rRNA and sequenced with the NGS ILLUMINA HISEQ and process like trimming and OTU clustering was done by the software like MOTHUR and PyNAST and the rarefraction curve was made to show the species richness
