Chromatin immunoprecipitation and sequencing for 3 histone marks (H3K27ac, H3K27me3 and H3K4me1) was performed on livers of male and female mice with a combined loss of Ezh1 and Ezh2. The DKO mouse model used in these analyses is a global deletion of Ezh1 and hepatocyte-specific deletion of Ezh2, and is described in Bae WK et al, FASEB J. 2015 May;29(5):1653-62. doi: 10.1096/fj.14-261537. PMID: 25477280. Chromatin was isolated from frozen livers from 7 week old male and female wild-type and double knockout (DKO; Ezh1-/-, Ezh2-f/f_Alb-Cre) mice and their floxed (f) controls. Sonicated chromatin from 3-4 individual livers from each group was immunoprecipitated with the following antibodies: H3K27ac (Abcam Cat # ab4729, 3 μg antibody per 15 μg of sonicated chromatin); H3K4me1 (Abcam Cat # ab8895, 1.2 μg antibody per 15 μg of sonicated chromatin); H3K27me3 (Abcam Cat # ab6002, 2 μg antibody per 10 μg of sonicated chromatin); and normal rabbit IgG (Santa Cruz, Cat # sc-2027, 3 μg antibody per 15 μg of sonicated chromatin).
