Transcriptomic analysis of vitamin D-treated human umbilical cord tissue-derived mesenchymal stem cells that were differentiated for 7 days into the myogenic lineage MSCs isolated from human umbilical cord tissue (UCT) and grown in αMEM with 20% fetal bovine serum on Laminin and Collagen Type 1-coated tissue culture plates were switched to medium containing αMEM with 15% KnockOut Serum Replacement. After 24 hours, MSCs from each of the biological replicates were treated with 100nM vitamin D or vehicle control for 3 days, with fresh medium being added every day. This was followed by induction of myogenic differentiation in both vitamin D-treated and untreated samples by the addition of medium containing 5% horse serum, 0.1 µM dexamethasone, and 50 µM hydrocortisone. After 7 days, total RNA was isolated from vitamin D-treated and control samples by Trizol (SIGMA) and processed for RNA-seq.
