ASNA1 (also known as TRC40) is a key component of the tail-anchored (TA) protein targeting pathway. To investigate its role in the heart, we generated cardiomyocyte-specific ASNA1 conditional knockout (ASNA1 CKO) mice. RNA-seq analysis was performed on E14.5 embryonic heart to identify transcriptional changes caused by ASNA1 deficiency. Our results reveal that ASNA1 is essential for maintaining the stability of TA proteins and regulating vesicular transport in cardiomyocytes, which are critical for normal cardiac development and function. To investigate the role of Asna1 in cardiac development, we generated cardiomyocyte-specific Asna1 knockout mice (Asna1^fl/fl; Xmlc2-Cre^+/−) by crossing floxed Asna1 mice with Xmlc2-Cre transgenic mice. Ventricular tissues were collected from embryonic day 14.5 (E14.5) knockout and control littermates (n = 4 biological replicates per genotype). Total RNA was extracted, and RNA integrity was assessed prior to library preparation. Strand-specific mRNA libraries were prepared using the Illumina TruSeq Stranded mRNA kit and sequenced on the Illumina HiSeq 6000 platform to generate ~30 million paired-end reads (150 bp) per sample.
