Here we show that GemC1-deficiency in mice results in the development of severe hydrocephalus impaired cytoarchitecture and function of the SVZ neurogenic niche due to lack of mature ependymal cells. Furthermore, we have showed that radial glial cells lacking GemC1 expression are not able to undergo commitment towards the ependymal lineage and acquire a neural stem cell fate. Collectively our findings shed light to the molecular mechanisms regulating NSCs and ependymal cells fate choice suggesting GemC1 as a critical regulator of the balance between ependymal cell and NSC generation.
