Comparative gene expression analysis was employed for deciphering the major metabolic pathways in actively growing and stationary-phase cultures of Cuniculiplasma divulgatum S5. The analysis of data revealed differences in expression profiles that could not be investigated otherwise, e.g. through the shotgun proteome studies. This transcriptome study accompanies physiological and metabolic studies to pinpoint the key niche adaptation mechanisms and disentangle evolutionary trajectories within Thermoplasmata. Overall design: After cultivation of Cuniculiplasma divulgatum and samples have been collected after 3 days, corresponding to its mid-log growing phase, and afer 5 days, corresponding to its early stationary phase. The goal is to compare both samples and check when and which proteins are expressed regarding its surface (archeaellum, pili, etc), production of carotenoids and metabolism. RNA-seq experiments were performed to compare the differential expresion of genes among the two growing phases.
