RNA sequencing was performed on three biological replicates of 1) CART19 + vehicle, 2) CART19 + 1 uM vecabrutinib, 3) CART19 + 10 uM vecabrutinib. Overall design: T cells from normal donors were transduced with a replication-incompetent lentiviral vector expressing a second-generation chimeric antigen receptor (CAR) consisting of an anti-CD19 single chain variable fragment (FMC63) fused to 4-1BB and CD3ζ intracellular domains as previously described. CART19 were activated for 72 hours in the presence of vehicle, 1 uM vecabrutinib, or 10 uM vecabrutinib prior to isolating RNA.
