Germline mutations are the source of evolution and contribute substantially to many health-related processes. In this study, we use whole genome deep sequencing data from parents-offspring trios to examine the de novo point mutations (DNMs) in the offspring. We studied correlation between parental ages with de novo mutation rates. Inclusion and Exclusion Criteria for Test Group Inclusion Criteria for Mothers Other than being admitted to the hospital for pre-term birth, the mother is in good general health Pre-term birth includes pregnancies that end between 23 and 36 weeks Age: ≥ 18 years The study is not race-restricted Exclusion Criteria for Mothers Trauma responsible for induction of pre-term labor Transfusion- in the previous six months Thrombophilia Atypical cervix Previous surgery on the cervix Exposure to diethylstilbestrol (DES) History of alcohol abuse during pregnancy History of drug abuse during pregnancy Weight extremes (anorexia, morbid obesity) Body Mass Index (BMI) less than 15 or greater than 35 Known exposure to hazardous chemicals (for example: dioxins) Known chromosomal disorders or genetic disease Cancer Inclusion Criteria for Fathers Good general health Age: ≥ 18 years The study is not race-restricted Exclusion Criteria for Fathers Non-paternity (if discovered through genotyping, family will be silently dropped from the study as described in the informed consent) Alcohol abuse 3 months prior to date of conception Drug abuse 3 months prior to date of conception Exposure to hazardous chemicals 3 months prior to date of conception Known chromosomal abnormalities or genetic disease Cancer Inclusion Criteria for Neonates Babies who are born between 23 and 36 weeks gestation Exclusion Criteria for Neonates Transfusion Known chromosomal or genetic defect Gross anatomic defect Inclusion Criteria for Multigenerational Family Members Good general health Age: ≥ 18 years The study is not race-restricted Positive family history of preterm birth Exclusion Criteria for Multigenerational Family Members Non-paternity (if discovered through genotyping, family will be silently dropped from the study as described in the informed consent) Inclusion and Exclusion Criteria for Control Group Inclusion criteria for mothers Mother is in good general health Delivery occurs after 36 weeks gestational age Age: ≥ 18 years The study is not race-restricted Exclusion criteria for mothers Transfusion- in the previous six months Thrombophilia Atypical cervix Previous surgery on the cervix Exposure to diethylstilbestrol (DES) History of alcohol abuse during pregnancy History of drug abuse during pregnancy Weight extremes (anorexia, morbid obesity) Body Mass Index less than 15 or greater than 35 Known exposure to hazardous chemicals (for example: dioxins) Known chromosomal disorders or genetic disease Cancer Inclusion criteria for fathers Good general health Age: ≥ 18 years The study is not race-restricted Exclusion criteria for fathers Non-paternity (if discovered through genotyping, family will be silently dropped from the study as described in the informed consent) Alcohol abuse 3 months prior to date of conception Drug abuse 3 months prior to date of conception Exposure to hazardous chemicals 3 months prior to date of conception Known chromosomal abnormalities or genetic disease Cancer Inclusion Criteria for Neonates Babies who are born after 36 weeks gestation Exclusion Criteria for Neonates Transfusion Known chromosomal or genetic defect Gross anatomic defect Vulnerable and Excluded Populations Vulnerable Neonates are considered to be a vulnerable population as they do not have the cognitive ability to understand the implications of participating in this study and do not have the ability to communicate their willingness to participate in the study. Thus informed consent is obtained from the mother. Excluded HIV infected populations are excluded because of the use of ARV drugs which could potentially be identified as the cause of pre-term birth. Populations exposed to hazardous chemicals to include environmental exposure, alcohol, illegal drugs, and OTC medications that represent a risk to mother or fetus which could potentially be identified as the cause of pre-term birth. Parents and neonates with chromosomal abnormalities or identifiable gene defects as these conditions could potentially be identified as the cause of pre-term birth. ]]>
