The goal of this study is to survey transcriptional machinery in intestinal stem cells from small and large intestine of mice. We established intestinal stem cell specific gene-regulatroy network to understand regional identity. Overall design: Small intestine (2-3cm from pylorous and 2-3cm from cecum) and large intestine (cecum to rectum) of mice aged 6-12 weeks were extracted, and opened horizontally and washed with cold PBS buffer. Small intestinal stem cell (Lgr5-EGFP+/CD24-) and large intestinal stem cell (Lgr5-EGFP+) were obtained from enriched intestinal crypts by FACS analysis. Sorted cells were directly applied with TRIzol™ Reagent (invitrogen 15596026) or ATAC lysis buffer.
