Recent studies have brought contrasting results concerning 6-methyl-adenosine (6mA) as an epigenetic mark in eukaryote genomes. Here we investigated the genomic characteristics of 6mA in drosophila larval central nervous system in wild type and tet mutant contexts using PacBio SMRT-seq. In parallel, we performed RNA-seq experiments to assess gene expression in these two conditions. Overall design: Drosophila melanogaster w1118 and tet null third instar larvae were raised on standard medium and their central nervous system was dissected at the third instar larval stage. High molecular weight genomic DNA was prepared and analyzed by SMRT sequencing using PacBio sequencing platform. Total RNA was extracted and analyzed by deep sequencing using Illumina HiSeq sequencing platform.
