Enamel mineralization is a tightly regulated process that depends on the coordinated activity of ameloblast-lineage cells (ALC). YAP, a key effector of the Hippo signaling pathway, has been implicated in tooth development, but the role of its post-translational modification in ameloblast mineralization remains unclear. To investigate transcriptional changes associated with Yap methylation during mineralization, ALC cells transduced with control lentivirus or a methylation-mimetic Yap mutant were subjected to mineralization induction for 7 days. RNA sequencing was performed to compare global gene expression profiles between these two conditions. Differential gene expression and pathway enrichment analyses revealed transcriptional changes related to mineralization, with prominent enrichment of mitophagy and PI3K/Akt signaling pathways. Collectively, this dataset provides a basis for further investigation of Yap-methyation regulatory mechanisms during ameloblast mineralization.
