We developed methods that reproduce the osteogenesis process using human iPS cells and addressed gene expression dynamics in the differentiation by single cell RNA-sequencing. We constructed a 3D culture system with type I collagen gel, and observed migration of the cells into the gel matrix. We collected two samples: (i) cells migrated into the matrix, and (ii) whole cells including both cells on the surface of the gel matrix and cells migrated into the matrix. We compared the two samples to analyze gene expression traits related to the differentiation.
