Marine cyanobacteria are well-known as a rich source of structurally intriguing chemical entities to serve as therapeutic leads addressing unmet medical needs, especially in the field of fighting against cancer. Lagunamide D, a novel macrocyclic depsipeptide was identified from cyanobacteria with single-digit nanomolar antiproliferative activity against multiple cancer cell lines. The goal of this study was to explore the mechanism of action of this potent molecule to enable in-depth understanding of its roles in modulating cellular processes. RNA-sequencing analysis was applied to get an unbiased global view of how lagunamide D function in cells and reveal its actions on transcriptome scale at early (3 h) and late (12 h) time point.
