We previously found that the downregulation of METTL14 played a crucial role in maintaining breast cancer stem-like properties. The present study aims to explore the impact of METTL14 knockdown on gene expressions and m6A modifications related to stemness in order to elucidate the underlying mechanisms. Overall design: To investigate the underlying mechanism through which METTL14 governs the stemness of breast cancer, we generated MCF7-shM14 cell lines wherein the expression of METTL14 was knocked down by shRNA. We conducted MeRIP-seq (m6A-seq) and RNA-seq analyses on these cells, comparing them to the control cells (MCF7-shNC).
