In the present study we used transcriptome sequencing to study the gene changes in the development of vegetative propagation in two Eucalyptus cultivars - DH201-2 (high capacity of vegetative propagation) and GL9 (low capacity of vegetative propagation). We identified some genes related to the callus development and the capacity of vegetative propagation, such as auxin related genes, embryogenesis related genes, ethylene related genes, ribosomal protein genes, histone genes, heat shock protein (HSP) genes and various TFs. Further, we identified the variants in these two cultivars that might trigger the vegetative propagation in DH201-2. This is the first time to study the transcriptome of vegetative propagation in Eucalyptus. Our results will improve our understanding about the molecular mechanisms during the vegetative propagation and the genes controlling vegetative propagation ability. The output of this study will also provide a valuable resource for future studies and will benefit the research in this field and the breeding program of Eucalyptus.
