We analyzed coding and noncoding transcript abundance in primary differentiating brite adipocytes derived from murine inguinal white adipose tissue, 24 hours or 72 hours in response to lncRNA Ctcflos knockdown at day 1 of differentiation Overall design: We prepared primary preadipocytes from stromal-vascular fraction of murine inguinal white adipose tissue of 129SvEv mice, induced them at sub-confluence for 2 days and differentiated them for 1 day in the presence of rosiglitazone to induce browning. Cells were reverse transfected with lncRNA Ctcflos-targeting or non-targeting control LNA Gapmer Antisense oligonucleotides [50nM] and harvested either 24 hours or 72 hours post transfection. We sequenced total RNA and determined transcript abundance.
