Transcription factors of the ZBTB family play critical roles in various physiological and pathological processes. Determining the DNA-binding motifs of these transcription factors is essential for understanding their regulatory functions. Although the binding motifs of many ZBTB family members, such as ZBTB6, ZBTB7A, ZBTB11, ZBTB12, ZBTB14, ZBTB17, ZBTB18, ZBTB20, ZBTB21, ZBTB22, ZBTB24, ZBTB26, ZBTB32, ZBTB33, ZBTB37, ZBTB40, ZBTB42, ZBTB43, ZBTB44, ZBTB45, and ZBTB48, have been experimentally characterized, the binding motif of ZBTB5 remains unidentified. To address this gap, we performed ZBTB5 CUT&RUN-Seq in the 293T cell line to uncover and validate the DNA-binding motif of ZBTB5, thereby facilitating further functional studies of this transcription factor.
