Human B-ALL cells become chemoresistant to multiple chemotherapies if exposed to adipocyte-secreted factors. To identifiy gene alterations underlying adipocyte-mediated chemoresistance, we performed RNA-sequencing analysis on untreated human B-ALL cells and those treated with the chemotherapy metotrexate. Human B-ALL cell lines (n=2; REH and RCH-AcV) were cultured in unconditoned, bone marrow stromal cell, or adipocyte-conditioned medium (CM) for 48 hours or pretreated with CM for 24 hours followed by 24 hours of methotrexate treatment. All samples were collected after 48 hours of the initial plating and process for RNA-sequencing.
