Comparative transcriptomic analysis of primary mature adipocytes from normal mice, obese mice, and obese mice undergoing B2M adipocyte-specific knockout

Chronic low-grade adipose inflammation caused by the dysregulated interaction between adipose-resident immune cells and hypertrophic adipocytes is involved in the obesity-related metabolic disorders, but the underlying mechanisms remain incompletely elucidated. In this study, we conducted a comparative transcriptome analysis of mature adipocytes purified from epididymal adipose tissue (epiWAT) of lean mice fed a normal diet for 16 weeks and obese mice fed a high-fat diet for 16 weeks. Our findings suggest that B2-microglobulin (B2M) may serve as a critical mediator linking obesity to adipocyte inflammation. To further investigate this hypothesis, we generated adipocyte-specific B2M knockout (cKO) mice and performed transcriptomic profiling of mature adipocytes isolated from epiWAT in these cKO mice after 16 weeks of high-fat diet feeding.
