This study investigated the bacterial communities present in human breast milk under three different preparation methods such as fresh and freeze-dried (lyophilized); and attempted to increase the microbial diversity by combining the freeze-dried samples. Milk samples were collected from 13 mothers and analyzed as fresh, individually freeze-dried, and as combination of freeze-dried from multiple donors. The microbial diversity across these sample types were compared using 16S rRNA gene sequencing. The combned samples depicted higher diversity and a balance of bacterial species in comparison to individual samples. The samples showed higher representation of common commensal organisms such as Streptococcus salivarius and Lactococcus raffinolactis. These findings highlighted the impact of sample processing on microbial composition of human milk and a brought to limelight a strategy to increase microbial diversity without the use of external compositions.
