A multifunctional population of Vg4 T cells are inhibited by Yersinia pseudotuberculosis. Mesenteric lymph node (MLN) suspensions from L. monocytogenes infected mice were stimulated with 10 MOI of WT or YopJ^C172A Y. pseudotuberculosis for 24 hours. Antibiotics were given 2 hours post-stimulation. Five hundred Vg1.1/2- CD44hi CD27- gd T cells from each stimulation were flow sorted and processed for RNA sequencing. In a separate experiment, MLN suspension from L. monocytogenes infected mice were stimulated with 1 MOI of WT Yptb-bla (Yop translocation reporter). Five hundred Yop+ or Yop- Vg1.1/2- CD44hi CD27- gd T cells were flow sorted and processed for RNA sequencing.
