Example of a photoconversion experiment on XP4PA cells transiently expressing low levels of XPC-mOrange. Cells were locally irradiated (5 m, 100 J/m2) and monitored in the orange channel (543 nm; non-photoconverted) and the far-red channel (633 nm; photoconverted). 30 min after local UV irradiation, the local accumulation of XPC-mOrange was photoconverted with 488 laser light. The levels of non-photoconverted XPC-mOrange increased at the local damage, while the levels of photoconverted XPC-mOrange decreased due to the rapid exchange of XPC at the damaged site.
