Trained immunity is the phenomenon whereby innate immune cells such as monocytes or macrophages undergo functional reprogramming after exposure to certain microbial components, diets, or metabolites, altering their responses to future exposures. Trained immunity is observed in hypercholesterolemia, dislipidemia, and in response to high fat diet. In this study monocytes from obese adolescents and controls were exposed to microbial stimulation in vitro or media alone, and transcriptional profiles were generated. Overall design: Blood was collected using venapuncture and PBMCs were isolated and viably frozen. Upon analysis, PBMCs were thawn, viable monocytes isolated with negative isolation and monocytes were stimulated for 4 hours with medium alone or purified LPS (TLR4 ligand). RNA from purified monocytes was isolated and the RNA-seq data was analysed. Comparisons for differentially expressed genes were made using either the unstimulated sample as reference (for stimulated vs unstimulated) or controls (for adolescents with obesity vs controls)
