Land-use change can significantly alter the wetland methane (CH<sub>4</sub>) cycling process, thereby affecting ecosystem services of wetlands. However, the impact of land-use on methanogens, methanotrophs, and CH<sub>4</sub> fluxes in karst wetlands remains unclear. The objective of this study is to compare CH<sub>4</sub> emissions, methanogens, and methanotrophs in a lake wetland (LW) and its adjacent paddy fields (PF) and abandoned paddy fields (PF-A) in Huixian karst wetland ecosystem and explore the mechanisms by which microorganisms and environmental factors caused by land-use change drive CH<sub>4</sub> emissions. The topsoils (0-20 cm) from LW, PF, and PF-A were collected. The CH<sub>4 </sub>fluxes were measured using static-dark-chamber combined gas chromatography. The abundance, diversity, and composition of methanogens and methanotrophs were detected with quantitative PCR (qPCR) and high-throughput sequencing, respectively.
