Uncoupling of DNA cleavage and repair during programmed genome rearrangements jeopardizes assembly of the somatic genome in Paramecium The present study aims to test this assumption, by addressing a simple question: what would be the consequences of uncoupling the DNA cleavage repair steps of the reaction during rearrangements? This will correspond to restore the bona fide situation of repair following accidental DSBs, where breaks will be introduced first, and then the recruitment of repair proteins will occur in response to the presence of DNA ends. How will the quality of the junctions, and more generally the quality of programmed genome rearrangements be affected in these conditions?
