The embryo of B. mori, an important economic and model insect of Lepidoptera, was demonstrated that its development was affected by DNA methylation. In this study, we found that DNA methylation enhances the expression of the hypermethylated gene zinc finger protein 615 (ZnF 615); Loss of Dnmt1 or ZnF 615 causes embryonic lethality. This study reveals a regulatory pathway of DNA methylation to insect embryonic development, that is intragenic DNA methylation enhances protein metabolism-related genes expression through upregulating the embryo-specific ZnF 615 to ensure the development from fertilized egg to multiple individuals (larvae).
