We observed three different matrix-producing variant colonies of Escherichia coli UTI89 population from the biofilm pellicle, when plated on congo red (CR) agar. One among which was a white colony variant (WCV), which displayed smooth and white (SAW) colony morphology on CR agar plate. The SAW morphology is seen when the cells in the colony does not produce the predominant biofilm matrix components like cellulose and curli. The other two variants that were found were called as red colony variant (RCV) and heterogeneous colony variant (HCV). In the study, we performed competition studies between the variants and found that the interactions are context dependent.
