Investigating the effect on CAR T cell phenotype by overexpressing FOXO1 Overall design: Murine C57BL/6 splenocytes were transduced to express a human HER2 directed CAR and a constitutively active variant of Foxo1 (Foxo1-ADA) or empty vector. CAR T cells were cultured with a CAR agonistic antibody for the HER2 CAR over 72 hours, purified and analyzed with 3' RNA seq.
