TIGER Predictions for all GENCODE v41 EEJ gRNAs. Table includes TIGER predictions for 2,255,040 EEJ gRNAs–tiling 8 gRNAs per EEJ across all GENCODE v41 basic annotation transcripts (protein-coding and lncRNA). ‘Chr’, ‘Start’, and “End’ are gRNA coordinates. ‘Gene ID’, ‘Gene Name’, and ‘Gene Type’ match GENCODE v41 and ‘Junc IDs’ and ‘Guide IDs’ were generated in Isoviz to offer consistent and unique IDs. The 23bp ‘Guide Sequence’ is the reverse complement of the targeted RNA–you will need to add appropriate overhangs for cloning into a plasmid. ‘TIGER score’ is a prediction of gRNA efficiency between 0 and 1, where a score > 0.2 is likely to be active. These predictions are from the original TIGER model (Wessels et al. 2023) and should be very similar to predictions on the TIGER webtool–the only difference is that the webtool doesn’t include ‘Cell non-specific set’ of features discussed in Figure 2. ‘EEJ Category’ labels whether the EEJ is common, fully unique, partially unique, or single gene unique as discussed in Figure 1 within GENCODE v41 annotations. ‘Transcript Isoforms’ shows which annotated isoforms each junction matches. ‘Target Sequence 50bp’ gives the Target RNA sequence–which is the reverse complement of the gRNA sequence–for -25bp to +25bp around each EEJ. Importantly, this is the sequence that can be copy and pasted into the TIGER webtool to look for any gRNA off targets. ‘Filtered’ column provides additional consideration for whether a gRNA should or should not be used. Most rows are missing information here but some include Likely Off-target or Homopolymer as our own rationale for filtering them from testing or analysis.
