Reduced NotchR signaling has been implicated in the reduced division and differentiation of stem cells in a number of tissues. The Protein-O-fucosyltransferase 1 gene (Pofut1) encodes a glycosyltransferase that O-fucosylates EGF-repeat-containing proteins, including Notch Receptors (NotchRs), and absence of Pofut1 glycosylation on NotchRs inhibits ligand-induced NotchR signaling. Here, we have deleted Pofut1 in the brain by making NestinCrePofut1FF mice to investigate the role of loss of Pofut1 expression on self-renewal and differentiation of adult neural progenitor cells (ANPs). Overall design: A total of six samples in three groups were analyzed for this study. The first group is two replicates of Nestin cre/Pofut1flox/flox brains. The second group is two replicates of Nestin cre/Pofut1 wild type brains. The third group is two replicates of Pofut1flox/flox brains.
