Needling on Trigger Point Promotes Muscle Regeneration after Bupivacaine Injection Induced Gastrocnemius Muscle Injury via RNA sequencing analysis

Purpose: We used RNA sequencing to investigate the effect of acupuncture treatment on regeneration after myotoxic muscle injury Methods:Methods:Gastrocnemius muscle mRNA profiles of 6-7 months-old WT(wild-type), Sham(Saline injeciton), Injury(0.5% Bupivacaine injecion) and Acup(1-time needling 24 hour after bupivacaine injection site) mice were generated by deep sequencing, in triplicate, using Illumina Hiseq platform. The sequence reads that passed quality filters were analyzed with DESeq2 R package to identify differently expressed genes with padj<0.05. qRTâPCR validation was performed using TaqMan and SYBR Green assays Results: Needling on trigger point once could change genetic expressions. Acupuncture once up regulated 623 genes and down regulated 65 genes compared to Injury muscle. Acupuncture 24 hours after injury promotes muscular regeneration. Conclusions: Our research first reported that 1-time needling 24 hours after muslce injury can promote muslce regeneration which will provide novel therapeutic targets for treatment of myofasical pain syndrome Overall design: Gastrocnemius muslce mRNA profiles of 6-7 months-old mice were generated by deep sequencing, in triplicate, using Illumina Hiseq.
