We aim to gain insight into the competitive adsorption behavior of monoclonal antibodies (mAbs) and a non-ionic surfactant (Poloxamer 188) at a liquid-hydrophobic solid interface. This is representative of interfaces that mAbs will encounter during pharmaceutical formulation, transport, and storage. Upon adsorption and desorption at interfaces, mAbs can irreversibly aggregate, reducing drug efficacy and shelf-life. The understanding of mAb stability is still elusive. Here, we plan to utilize neutron reflectometry to understand the adsorption behavior of mAbs with different long-term stabilities, to probe the ability of a Poloxamer 188 to shield a hydrophobic interface from mAbs, and to gain insight into the effect of order of addition of each component. This experiment will have a high industrial impact, and may be able to assist in the optimization of new therapeutic formulations.
