Single-cell transcriptome profiling of radial glia cells in zebrafish adult telencephalon in response to traumatic brain injury

In zebrafish brain, radial glia cells (RG) show a remarkable ability to regenerate damaged neural tissue by reentering cell proliferation and produce neural precursors. To understand how RG respond to brain damage and initiate the regenerative response, we applied single-cell transcriptome analysis to RG in zebrafish adult telencephalon. Overall design: Radial glia cells were isolated from the telencelphalon of adult zebrafish transgenic line Tg[GFAP:dTomato]nns17Tg by Fluorescence-activated cell sorting (FACS). Collected dTomato+ radial glia cells were subjected to single cell RNA-seq analysis using Chromium Single Cell 3' Reagents Kits v3 (10x chromium). The sequencing data were analyzed using 10x Genomics Cell Ranger 3.1.0 with default settings and zebrafish reference genome (GRCz11).
