ROF1 induction experiments [Agilent]

Measure global transcriptional responses to ROF1 induction in S288c budding yeast Experiments with phosphate-limited chemostats were performed as described in Hackett et al (MSB, 2020). Extraction, labeling, and hybridization of RNA were performed as described in Hackett et al (MSB, 2020). Microarrays were imaged using an Agilent SureScan G4900DA Microarray Scanner. Images were processed using Agilent Feature Extraction software and a custom R script. Fluorescence intensities were floored at a value of 50. The resulting data were hierarchically clustered (Pearson distance metric with average linkage) using Cluster 3.0 and visualized in R using the pheatmap package.
