Identification of homoeologous single nucleotide polymorphisms in Brachypodium hybridum

We sequenced an allopolyploid grass Brachypodium hybridum and its ancestor Brachypodium stacei to identify their sequence differences between homoeologs. Genomic DNA from leaf tissues of B. hybridum and B. stacei were extracted by using the DNeasy Plant Mini Kit (QIAGEN). Libraries for single-end DNA sequencing were obtained by using the Ion Xpress Plus Fragment Library Kits (Life Technologies). The sequencing analysis was performed using the Ion Proton sequencer (Life Technologies).
