DNA barcoding is a powerful technology that can be used to rapidly test efficiency of delivery of many different delivery vehicles, such as LNPs, at the same time. However, DNA barcodes are small oligonucleotides and may not form LNPs with the same characteristics as those formulated with larger mRNA transcripts. We tested the relative delivery of LNPs formulated with DNA barcodes and +/- co-encapsulated mRNA by pooling these LNPs, injecting into mice, and testing for variability in delivery to different organs.
