To identify polyA+ transcripts that were differentially expressed in MCF10A human breast epithelial cells or hTERT RPE-1 retinal pigment epithelial cells following treatment with MIR-28 family member microRNA mimics, we profiled cellular RNAs by Illumina sequencing. Treatment conditions were a non-targeting siRNA (siNT, negative control), hsa-miR-28-5p microRNA mimic, or hsa-miR-708-5p microRNA mimic (Horizon Discovery/Dharmacon). Cells were seeded at 100,000 cells/well in 6-well plates for 24 h, then transfected with RNAi reagent for 72 h. Total RNA was harvested with TRIzol and purified. The Yale Center for Genomic Analysis prepared 100 bp paired-end libraries for sequencing after polyA+ (oligo dT) selection, and conducted the Illumina sequencing experiments.
